Antisense Therapy and the creation of the SOD1 protein.Jan 29, 2012 Visit sam-fernandez.com. A project I created for the Muscular Dystrophy Association. It deals with the concept of Antisense therapy - in which scientist kill targeted strands of RNA that are responsible for the creation of the SOD1 protein. The animation was created before the script and voice over, so there is a bit of a disconnect between what's happening on the screen and what's being said. This is not a finished product. It was put on hold indefinately and never fully realized. The animation itself was shown during the Jerry Lewis 2010 telethon, which is broadcast nationally, during several of the research updates.

Bidirectional Transcription and Antisense Repression of ataxin-7Nov 10, 2011 CAG repeat expansions in the ataxin-7 gene cause the neurodegenerative disorder SCA7. Al La Spada and colleagues have discovered that ataxin-7 expression is governed by SCAANT1, an antisense non-coding RNA transcribed from an adjacent alternative promoter. Transactivation of SCAANT1 is linked to repression of ataxin-7 transcription and chromatin remodeling, suggesting that altered epigenetic regulation may contribute to disease pathology. Dance your PhD 2011Feb 08, 2012 Our entry for this year's "Dance your PhD" contest, titled "Antisense Oligonucleotides-Mediated Exon Skipping for Duchenne Muscular Dystrophy". See gonzolabs.org for all entries. Duchenne muscular dystrophy (DMD) is a lethal muscle disease which is caused by non functional dystrophin protein. Normal dystrophin gives a proper structure to our muscle and enables us to perform physical activities, such as walking... and dancing! The dystrophin protein is encoded by the DMD gene, which consists of arrays of exons (E) and introns (I). To make dystrophin protein, the introns have to be spliced out by the spliceosome (pink). The exons are then joined together and the amino acids (AA) are synthesized. When there is a mutation (STOP) in the DMD gene, the dystrophin protein cannot be synthesized properly and therefore not functional. This non functional dystrophin makes the muscle weak, more prone to damage and eventually degenerate. Currently there is no curative treatment for this disease. In our lab, we are working on an approach called "exon skipping". We try to correct this faulty gene using a molecule called antisense oligonucleotides (AON, green). What does it do? The AON is designed to target, bind and mask/hide the mutated exon. The hidden exon will be treated as an intron by the spliceosome, and spliced out together with the other introns. Hence the AON is a "skip man" :) Dystrophin produced after exon skipping will be shorter, but should be sufficient for the affected ...
